Hello, I would appreciate recommendations for a release to be used in a PU mold, that will allow plaster to be cast, and painted with water based colors upon demolding.

[Try mold soap. I get it at the ceramic supply as a solid green gel, which can be thinned with alcohol to the desired consistency. Wash the castings in water to remove any residue before painting.]

do you need a release for plaster with a urethane mould? ..... I wouldn't have thought so

PU? By this do you mean a RIGID mold made of polyurethane resin? Or do you mean a flexible mold made of polyurethane elastomer?

If resin- try the mold soap- you can also buy it in liquid form from Perma-Flex mold co. or its distributors. When using mold soap ( which is actually a kind of wax) - the key is to allow it to dry completely before casting- and to lightly BUFF the soaped surface with a dry brush to bring up a gloss in the wax.

If it is elastomer, then you should not require any separation agent to cast plaster.

some elaastomers will exude an oily film that will cause 'fisheyes' in any paint you try to apply to the casting. This can be the result of poor mixing- but can be reduced or eliminated by throroughly de-greasing the inside of the mold surface prior to casting with a solvent based de-greaser- something that flashes off completely like acetone or methylene chloride.
